Research Overview

β-Carbolines are a family of tricyclic indole alkaloids found endogenously in human brain and cerebrospinal fluid as well as in plants (Peganum harmala), tobacco smoke and roasted coffee [1]. 9-Methyl-β-carboline is a synthetic, methylated derivative whose pharmacology departs sharply from the parent norharman: rather than the MAO-A-inhibitory profile of unmethylated β-carbolines, 9-Me-BC exhibits a complex but reproducible profile of dopaminergic neuroprotection, neurogenesis and anti-inflammatory activity in primary mesencephalic neuron cultures and in vivo [1,2]. Mechanistic work has identified upregulation of GDNF, BDNF and tyrosine-hydroxylase expression, suppression of microglial activation, and increased proliferation of dopaminergic progenitors as core features [2]. In MPTP and 6-OHDA rodent models of Parkinson’s disease, oral 9-Me-BC at clinically relevant doses partially restores striatal dopamine, rescues motor performance and induces measurable hippocampal neurogenesis [3]. The compound has emerged as one of the most distinctive research tools available for combined neuroprotective-neurogenic pharmacology — a profile not replicated by any approved neurodegenerative-disease drug.
