Research Overview

DSIP (delta sleep–inducing peptide) is a neuropeptide nonapeptide investigated in experimental systems for its roles in sleep–wake regulation, circadian biology and neuroendocrine modulation, including stress-adaptive signaling pathways.

Primary Research Areas

  • Sleep regulation & circadian rhythm: models of slow-wave sleep, sleep architecture and biological clocks [7].
  • Neuroendocrine & HPA-axis signaling: research on hypothalamic–pituitary–adrenal interactions and stress adaptation [3].
  • Stress-response models: experimental work on acute and chronic stress load and resilience mechanisms [8].
  • Nociception & sensory processing: exploratory studies on pain perception and neuronal signaling circuits [2].
  • Alcohol and opioid withdrawal: 107 inpatients withdrawing from alcohol or opiates were given DSIP into a vein; staff judged the withdrawal signs cleared or improved in 87% of the alcohol group and 97% of the opiate group. There was no control group and no blinding in this 1984 series, so it records what happened, not what caused it [1].
  • Low mood alongside long-standing pain: seven patients with migraine, vasomotor headache, chronic tinnitus or psychogenic pain attacks were given DSIP into a vein on five consecutive days; pain fell in six of the seven and the depressive states that came with it lifted at the same time. Seven people, no placebo group, each compared with their own earlier history [2].
  • Growth hormone unchanged: eight healthy women were infused with DSIP at 25 micrograms per kilogram and their growth hormone and prolactin did not budge at rest, across the day, or when the system was pushed with arginine. It argues against the common claim that this peptide drives growth hormone release [3].
  • Seizures in rats: in rats given repeated sound-triggered seizures with metaphit, DSIP at 1 mg/kg reduced how often the seizures happened and how long they lasted. In a second study the peptide did nothing on its own but let an otherwise useless dose of valproate hold seizures off for six hours. Rats, injected, in one unusual seizure model [4,5].
  • Heart muscle during a heart attack: rats given 20 micrograms per kilogram of delta-sleep peptide an hour before an experimental heart attack kept much more of their normal heart-muscle stretch and squeeze than untreated rats. A 1985 rat study using pre-treatment, which is the opposite of how a person would ever encounter it [6].
